Determination of Purity of Food Colours using Paper Chromatography

Introduction

Food colours are substances added to food to enhance its appearance, compensate for colour loss during processing, or provide colour to colourless foods. Ensuring the purity of these additives is critical for consumer safety. Paper chromatography is a versatile, cost-effective analytical technique used to separate and identify components within a mixture, making it an ideal method for testing food dyes.

Principle of Paper Chromatography

Paper chromatography operates on the principle of partition chromatography. It involves a stationary phase (cellulose paper) and a mobile phase (a solvent or mixture of solvents). As the solvent moves upward through the paper via capillary action, it carries the components of the dye mixture with it. The rate at which each component travels depends on its relative affinity for the stationary phase versus the mobile phase. If a food dye is pure, it will move as a single spot. If it contains impurities or is a mixture, multiple spots will appear at different positions on the chromatogram.

Materials Required

  • Chromatography paper (Whatman No. 1 filter paper)
  • Food colour samples (e.g., commercial food dyes)
  • Solvent system (e.g., a mixture of water and ethanol or dilute acetic acid)
  • Chromatography chamber (a glass jar or beaker)
  • Capillary tubes for spotting
  • Pencil and ruler
  • Distilled water

Experimental Procedure

  1. Preparation: Cut a rectangular strip of chromatography paper. Draw a thin pencil line approximately 2 cm from the bottom edge.
  2. Spotting: Using a clean capillary tube, apply a small, concentrated spot of the food colour sample onto the pencil line. Allow it to dry, and repeat to ensure the spot is concentrated.
  3. Development: Pour the solvent into the chromatography chamber to a depth of about 1 cm. Ensure the solvent level is below the pencil line.
  4. Running: Suspend the paper strip in the chamber, ensuring the spot is above the solvent level. Cover the chamber to saturate the atmosphere with solvent vapor.
  5. Completion: Allow the solvent to rise up the paper until it reaches near the top. Remove the strip and mark the solvent front with a pencil.
  6. Drying and Analysis: Allow the paper to air dry. Observe the number of spots present and measure the distance traveled by each component from the baseline.

Analysis and Interpretation

The purity of a food colour is determined by calculating the Retardation Factor (Rf value):

Rf = (Distance traveled by the solute) / (Distance traveled by the solvent front)

A single, well-defined spot indicates a high level of purity. If multiple spots are observed, it suggests the sample is a mixture of dyes or contains impurities. By comparing the Rf values of the samples with known standards, specific dyes can be identified and verified against regulatory safety lists.

Paper chromatography provides a simple yet effective way to assess the quality and identity of food dyes. While more advanced techniques like HPLC (High-Performance Liquid Chromatography) are used for quantitative analysis in industrial settings, paper chromatography remains a fundamental educational and preliminary screening tool to ensure that food additives meet the necessary standards for human consumption.
