Standard Protocol (25minute workflow)

  1. Deparaffinize slides in the provided xylene substitute (22min).

  2. Hydrate through graded ethanol (100%, 95%, 70%, each 30s) and rinse in distilled water.
  3. Stain nuclei with Hematoxylin for 5min. Rinse briefly in running tap water.
  4. Blueing in Scotts tap water for 30s to develop the distinct bluepurple nuclear hue.
  5. Eosin staining for 23min, depending on desired cytoplasmic intensity. Rinse in distilled water.
  6. Dehydrate through 95% ethanol (30s) and 100% ethanol (230s).
    Optional: a quick dip in xylene substitute to clear the tissue.
  7. Mount using the provided resin and place a coverslip. Allow to set for at least 5min before imaging.
