Proteomics Facility: Sample Submission Procedure
The Proteomics Facility is dedicated to providing high-quality mass spectrometry analysis for biological research. To ensure the integrity of your data and the efficiency of our workflow, all users must adhere to the following standard submission procedures.
1. Project Consultation
Before submitting samples, new users are required to schedule a consultation with the facility manager. This ensures that the experimental design is optimized for mass spectrometry and that we have the necessary resources to meet your research goals. During this meeting, we will discuss sample preparation methods, protein concentration requirements, and anticipated turnaround times.
2. Sample Preparation Guidelines
Quality control begins at the bench. Because mass spectrometry is highly sensitive, sample purity is paramount. Please adhere to the following preparation standards:
- Contamination Prevention: Avoid the use of detergents (e.g., SDS, Triton X-100), polymers (e.g., PEG), or excessive salts. These chemicals interfere with ionization and can damage sensitive mass spectrometer components.
- Protein Concentration: Samples should be provided in clean, low-protein-binding microcentrifuge tubes. Ensure that protein concentrations are measured using a standard assay (e.g., BCA or Bradford) before submission.
- Labeling: Every tube must be clearly labeled with a permanent marker. Ensure labels correspond exactly to the digital submission form provided to the facility.
Pro-Tip: Use only high-purity LC-MS grade solvents if you are performing in-house digestion. If you are submitting gel bands, ensure they are excised using a clean, new scalpel blade and placed into clean tubes.
3. Submission Documentation
Each project must be accompanied by a completed Sample Submission Form. This digital form contains critical metadata, including:
- Principal Investigator (PI) information and billing/grant account codes.
- Sample identification (e.g., strain names, treatment conditions, cell types).
- Buffer composition (e.g., pH, concentration of additives).
- Expected protein quantities per sample.
- Specific analytical goals (e.g., identification, quantification, or post-translational modification mapping).
4. Logistics and Sample Delivery
Once your project is approved and the submission form is finalized, please coordinate your sample delivery:
- Temperature Requirements: All samples must be shipped or hand-delivered on dry ice to maintain protein stability. We do not accept samples that have undergone multiple freeze-thaw cycles.
- Timing: Please inform the facility via email 24 hours prior to your expected delivery time.
- Storage: Upon arrival, samples will be verified against the submission manifest and stored in a -80C freezer until analysis begins.
5. Quality Control and Data Processing
Upon receipt, the facility will perform a preliminary quality control assessment. If a sample is deemed insufficient or highly contaminated, you will be notified immediately to discuss potential remedial actions. Following the mass spectrometry run, you will receive a notification containing a link to your raw data and a summary report of the findings. Please note that data is archived for a period of six months; users are encouraged to download their files promptly after completion of the project.
6. Acknowledgment Policy
The Proteomics Facility relies on the recognition of our contributions to facilitate continued funding and service. We request that users acknowledge the facility in all publications, posters, or presentations resulting from data generated by our services. Proper acknowledgment is vital for our facility's sustainability.
